Abstract
Roughly a third to a half of all newly discovered drug candidates run into the same wall during early Development: they simply will not dissolve in water fast enough, or in high enough amounts, to reach the Bloodstream in a therapeutically useful way. Pharmaceutical scientists have tried almost everything to get around this problem salt formation, co-solvents, cyclodextrin complexation, lipid carriers, amorphous solid Dispersions and each of these has its place. But one approach has quietly become the workhorse solution for a surprisingly large number of marketed products: shrinking the drug crystal itself down to a size between roughly ten and a thousand nanometres, without diluting it in any carrier. This is the nanocrystal, or “Nano suspension,” Approach, and it has moved from a laboratory curiosity in the early 1990s to a technology that underpins several Billion-dollar drug products and a growing list of applications well beyond the oral route.
